Metagenome Techniques for Detection of Pathogens Causing Ocular Infection

Metagenomic analysis is the comprehensive study of DNA using clinical specimens organisms including bacteria, fungi, and viruses. In this study, we investigated efficacy metagenomic for diagnosing ocular infections, 11 keratitis cases, four iridocyclitis one endophthalmitis case. Corneal scraping, aqueous humor, vitreous were collected respectively. Ocular used bacterial fungal culture, PCR detecting viral DNA. Shotgun sequencing 150 bases single end was performed by Illumina MiSeq® System. Sequence retrieved from database at NCBI a MegaBLAST search. Since Propionibacterium spp. are commensal bacteria found surface, they excluded analysis. Six cases (37.5%) positive culture or PCR. Metagenome techniques revealed that 9 (56.3%) included genomes considered pathogenic in specimens. Five (31.3%) possessed like themselves detected negative PCR, metagenome Moreover, pathogens (HSV-1, 2 cases; VZV, 1 case) only an sample can detect microbial genome comprehensively, pathogens, which not conventional examination.
